Determining the Mode of Inheritance in an Eye Color Mutation
>Select "New"
>Open Fruit Fly Exercise 2 - Level 1
You have been working with Drosophila melanogaster flies. By now, you are
familiar with wild-type flies and know that they have red eyes (wt), and
occasionally white eyes (WE). In one of your fly vials, you discover a male fly with
orange colored eyes. You are intrigued since you have never seen a fly with such
an unusual eye color. You call this mutant "Orangeye" (OE). You decide to do
some genetic analysis of this unusual mutant. To do this, you want to set up a
test cross between the Orangeye mutant and a wild-type fly.
Q1) What is the sex of the Orangeye (OE) Fly?.
Now that you know sex of your mutant, mate the mutant with a wt fly.
Q2) What is the percentage of each Phenotype of the F1 generation?
wt:
%
OE:
%
WE:
%
Q3) Using the results of your test cross, is the OE trait dominant or recessive to Red
Eye?
Q4) What are the possible genotypes for individuals in the F1 generation (use "A" for
dominant and "a" for recessive alleles?
To test your hypothesis, select one Male from the F1 generation, label it OEf2-M
and save it to your "Strains List". Next, select one Female from the F1 generation,
label it OEf1-F and save it to your "Strains List". Starting a new experiment, cross
the individuals from the F1 Generation.
Q5) What is the percentage of each phenotype by sex in the F2 generation?
wt-M:
%
wt-F:
OE-M:
%
OE-F:
%
Q6) What mode of inheritance could explain the phenotypic ratios observed in the F2
generation?
Q7) Draw a Punnett Square that could result in the phenotypic ratios seen in the F2
generation:
You share your unusual results with a friend who is also studying genetics in a
Drosophila lab. She tells you that she has a fly strain with another mutant eye
color, white. Your friend gives you a female fly, Whiteye (WE), from her true-
breeding white-eyed fly stock. She also tells you that the Whiteye mutant allele
confers a phenotype that is recessive to wild type.
In a new experiment, set up a cross between your OE mutant and the WE mutant.
Q8) What is the phenotypic ratio of the F1 generation (listed as wt:OE:WE)?
Q9) Do the results from the cross indicate that the mutations for OE and WE are on the
same gene or different genes?
To test your answer from Q8, set up a cross between the WE individual and a wt
individual. As was done for the OE test cross, select a male and female from the
F1 generation, label them WEf1-M and WEf2-F, and set up another cross between
the individuals from the F1 generation.
Q10) What is the percentage of each phenotype by sex in the F2 generation?
wt-M:
%
wt-F:
%
WE-M:
%
WE-F:
%
Q11) Do these findings support your answer from Q9? Why or why not?
