Transcribed Image Text:
The upstream region from GeneX is placed upstream of CAT in a Reporter Plasmid. A TxF named PREB is
known to affect GeneX transcription. PREB has 500aa and its gene is incorporated into a Producer Plasmid.
Two other Producer Plasmids are also constructed: aa1-400 and aa100-500.
Relative CAT activity is shown in the Table below for all three Producer Plasmids, as well as Reporter
Plasmids with various regions deleted..

Rationalize the results in the table.
YOUR ANSWER MUST INCLUDE A TABLE LIKE THE ONE BELOW.
1. You MUST give the name of the most likely cis elements in each of the ten regions (1-10), as they relate
to control of GeneX. Leave the table blank in any region where you do think there is NO cis element. At the
bottom of both answers, you may comment on some of your conclusions.

2. Draw a picture of PREB that looks like the one to the right and label the most likely structural elements in
each region. In the table also indicate where PREB binds.
Hint: It will be easiest to answer this question if you start your thinking at the top of a deletion column and
work down line-by-line (or start at the bottom and work up).
