In E.coli, the expression from most operons relies on sigma factor 70 (070). A subset of
operons relies on 032 which is expressed in place of σ70 in response to temp >42 °C
(heat shock). Gene expressions from these latter operons is necessary for the bacteria
to survive such a stressor. One or more of these genes also act through a positive
feedback loop in order to maintain σ32 expression.

To induce dsRNA expression against Gene X in your E.coli cultures, you grow them
overnight in an incubator set to 37 °C in media that contains the appropriate nutrient
signal. After you leave for the day, your lab mate who is growing a new thermophile
adds his cultures to the same incubator and turns up the temperature to that bacteria's
preferred 45 °C
In the morning, you proceed with your experiment, feeding your E.coli cultures to your
C. elegans and ultimately analyzing the resulting Gene X expression patterns using in
situ hybridization.
